This postdoctoral fellowship opportunity is offered jointly by the Purification Development and Bioanalytical Development Characterization departments within the Biologics Development organization.
Purification Development is responsible for development, scale-up, transfer and characterization of purification processes for early and late stage clinical and commercial manufacturing of therapeutic proteins and antibodies. In addition, the department develops next-generation commercial processes, provides technical expertise and support to improve ongoing clinical and commercial manufacturing process performance, evaluates new technologies, and develops platform technologies suitable for incorporation into clinical and commercial processes.
The Characterization group is responsible for the detailed analysis and characterization of proteins and antibodies from all stages of development. Detailed analysis of protein interactions, primary, secondary and tertiary structure, impurities, glycosylation and degradation pathways is performed by a multi-disciplinary team using state of the art analytical technologies. The results and expertise of this group play an important role in candidate selection, process development, understanding and assignment of critical quality attributes and establishment of structure-function relationships.
We are looking for candidates for the position of Post-Doctoral Fellow. This individual should be able to use scientific principles and professional practices to creatively and practically lead the execution of a cutting-edge research project bridging the disciplines of in silico purification process development and host cell protein proteomics. The candidate must ensure the highest standards of safety, environmental and regulatory compliance are maintained in the workplace.
Detailed literature review to assess the current state of the in silico process development and host cell proteomic areas of research
Development of a host cell protein database, including biochemical and biophysical properties of key host cell proteins and a comprehensive analysis of process removal capability of clinical and commercial processes
Development of first principles models using programming tools (i.e. MATLAB, Python) to simulate purification processes
Design and execution of high throughput experiments to evaluate, refine, and validate first principle models
Leveraging the principles of Design of Experiments (DoE) and statistical modeling, as appropriate
Design and execution of targeted LC/MS analyses to test specific hypotheses
Contribution to ongoing process development pipeline projects using host cell protein expertise gained throughout the post-doctoral training
Identification of collaboration opportunities across the Biologics Development organization and with partners in Research, Development, and Industrial Affairs
Publishing in first class technical journals
The candidate will receive extensive training in the experimental techniques and concepts of Purification Process Development, advanced analytical techniques including Mass Spectrometry, and experience modeling complex interactions. The opportunity provides unique cross-training in multiple biotechnology disciplines. The candidate will receive access to proprietary process knowledge, high-resolution proteomic data sets not typically available in academia, and real industrial feed streams for study. The disruptive improvements to our purification development approach resulting from this work should provide the candidate with an opportunity to contribute intellectually to the industry and the scientific literature.
Basic Qualifications :
Ph.D. from an accredited university in Chemical Engineering, Biochemical Engineering, Proteomics, Analytical Chemistry (Mass Spectrometry), Biochemistry, Biophysics, or related field
A strong understanding of first principles (mass balances, thermodynamics, kinetics) relevant to purification development
Ability to use programming tools (such as MATLAB, Python, etc.) to solve complex systems of differential equations
Ability to determine and develop approaches to complete milestones associated with specific projects, under general direction
Must have US work authorization
Experience with first principles modeling leveraging compiled coding languages (such as MATLAB, Python, etc.)
Experience in proteomic analysis using liquid chromatography-mass spectrometry (LCMS)
Hands-on experience with bench-scale protein purification, including clarification, tangential flow filtration, ultrafiltration/diafiltration, and column chromatography
Self-motivated, organized, attentive to details, and familiar with the relevant literature.
Able to thrive in a cross-functional working arrangement, open to vigorous scientific debate in a developing field of study
Excellent written and oral presentation and communication skills, including a desire and ability to realize external publications and presentations
